hey guys i have a 2000 diamante ls would like to know if i can reuse the intake gasket when i change the 3 plugs at the reare.

Welcome to MF Mentality.

As long as it's a metal gasket.  Chances are it is metal since you have to remove the plenum to get to the rear spark plugs.

Ill assume you mean plenum..  In which case, the answer is yes unless obviously damaged.

Yup, but...when there's two intake manifolds, one is called the plenum (or upper intake manifold) and the other (lower intake manifold) is called the lower intake manifold, lol.  When you say "intake manifold" in general, you could mean both the upper and lower, but when you say the plenum, we know that you mean the upper.  The upper needs to be removed to get to the plugs.  I took that at face value, Manybrews is trying to get you to be more specific for the others in here so that everyone understands the info that you're looking for [;)]. 
To add on to MB's comment, be careful when removing the plenum so that you don't bend the gasket as it may stick to the plenum...it may not, but be careful regardless.

How difficult is this task,to remove the plenum?

the plenum takes about 5 minutes to remove. look in the back part of it, there are 2 brackets that support the plenum and are attached by 4 bolts. not hard!!    greg....
